 ADVERSE EFFECTS ::

Sudden hypotension and bradycardia 

Cardiac failure 

Myocardial depression 

Bronchospasm in asthmatic patients 

Constipation nausea and vomiting 

Hypoglycemia and in masking of the symptoms of hypoglycemia 

Lethargy 

Mental depression and Fatigue 

Cold extremities 

Drugs acting on adrenergic nerves ::

(a) Guanethidine ::

It is a synthetic compound and produces blockade of adrenergic nerves it produces its action by depletion of noradrenaline from the sympathetic nerve terminals it produces fall in blood pressure lasting for 2-4 days it is well absorbed and slowly excreted and this gives a longer duration of action During therapy tolerance does not develop 

(b) a-Methyl Dopa ::

It prevents formation of noradrenaline from dopamine by inhibiting the enzyme beta hydroxylase thus inhibiting the biosynthesis of noradrenaline and consequently fall of blood pressure it shows reserpine like action by depleting noradrenaline from the brain it is effective by oral administration it is not suitable for malignant hypertension but is useful in moderate hypertension 

(c) Bretylium ::

It acts by inhibiting the release of noradrenaline by depressing the respose to nerve endings Thus it has no action on receptors it lowers normal as well as raised blood pressure it is not well absorbed Tolerance is developed during therapy This calls for an increase in dosage as treatment continues further

एड्रीनर्जिक नसों पर काम करने वाली दवाएं ::

(ए) गुआनेथिडीन ::

यह एक सिंथेटिक यौगिक है और एड्रीनर्जिक नसों की नाकाबंदी पैदा करता है यह सहानुभूति तंत्रिका टर्मिनलों से नॉरएड्रेनालाईन की कमी से अपनी क्रिया पैदा करता है यह 2-4 दिनों तक चलने वाले रक्तचाप में गिरावट पैदा करता है यह अच्छी तरह से अवशोषित होता है और धीरे-धीरे उत्सर्जित होता है और इससे लंबी अवधि मिलती है कार्रवाई चिकित्सा के दौरान सहनशीलता विकसित नहीं होती है

(बी) ए-मिथाइल डोपा ::

यह एंजाइम बीटा हाइड्रॉक्सिलेज़ को रोककर डोपामाइन से नॉरएड्रेनालाईन के निर्माण को रोकता है और इस प्रकार नॉरएड्रेनालाईन के जैवसंश्लेषण को रोकता है और इसके परिणामस्वरूप रक्तचाप में गिरावट आती है। मध्यम उच्च रक्तचाप में उपयोगी है

(सी) ब्रेटिलियम ::

यह तंत्रिका अंत के प्रति प्रतिक्रिया को कम करके नॉरएड्रेनालाईन की रिहाई को रोकता है, इस प्रकार यह रिसेप्टर्स पर कोई कार्रवाई नहीं करता है यह सामान्य के साथ-साथ बढ़े हुए रक्तचाप को कम करता है यह अच्छी तरह से अवशोषित नहीं होता है चिकित्सा के दौरान सहनशीलता विकसित होती है यह उपचार के रूप में खुराक में वृद्धि की मांग करता है आगे भी जारी है
