 PRAZOSIN ::

This is quinazoline derivative possessing a potent vasodilator action it has a specific a1-receptor blocking action however it appears that besides a1-blocking action it possess certain other properties that produce a potent vasodilator action it could be used alone in mild hypertension More often it is employed in combination with beta -adrenoceptor blockers or diuretics 

    It is well absorbed orally and metabolized in liver Half life is short (2-3 hours) but action persists for 10-12 hours Thus it is given in 0.5 mg dose twice a day 

Adverse effects include postural hypotension drowsiness and weakness it may cause tachycardia giddiness tiredness nausea diarrhoea palpitation and nervousness 

Minoxidil ::

It is a powerful arterial vasodilator acting by potassium channel opening with potentially serious side effects it is seldom used except in the therapy of resistant hypertension Major side effects are compensatory tachycardia pulmonary hypertension hypertrichosis headache pericardial effusion etc it also causes hair growth Thus minoxidil topical preparation (1-3%) are used to treat alopecia or baldness it causes increase in dermal blood flow and elongation of follicles in the area of alopecia

यह क्विनाज़ोलिन व्युत्पन्न है जिसमें एक शक्तिशाली वासोडिलेटर क्रिया होती है इसमें एक विशिष्ट a1-रिसेप्टर अवरोधक क्रिया होती है, हालांकि ऐसा प्रतीत होता है कि a1-अवरोधक क्रिया के अलावा इसमें कुछ अन्य गुण भी होते हैं जो एक शक्तिशाली वासोडिलेटर क्रिया उत्पन्न करते हैं इसे हल्के उच्च रक्तचाप में अकेले इस्तेमाल किया जा सकता है अधिकतर इसे बीटा-एड्रेनोसेप्टर अवरोधकों या मूत्रवर्धक के साथ संयोजन में नियोजित किया जाता है

यह मौखिक रूप से अच्छी तरह से अवशोषित होता है और यकृत में चयापचय होता है आधा जीवन छोटा (2-3 घंटे) होता है लेकिन क्रिया 10-12 घंटे तक बनी रहती है इसलिए इसे दिन में दो बार 0.5 मिलीग्राम खुराक में दिया जाता है

प्रतिकूल प्रभावों में आसन संबंधी हाइपोटेंशन उनींदापन और कमजोरी शामिल हैं यह क्षिप्रहृदयता, चक्कर आना, थकान, मतली, दस्त, घबराहट और घबराहट का कारण बन सकता है

मिनोक्सिडिल ::

यह संभावित रूप से गंभीर दुष्प्रभावों के साथ पोटेशियम चैनल खोलने द्वारा कार्य करने वाला एक शक्तिशाली धमनी वासोडिलेटर है इसका उपयोग शायद ही कभी प्रतिरोधी उच्च रक्तचाप के उपचार में किया जाता है प्रमुख दुष्प्रभाव प्रतिपूरक क्षिप्रहृदयता, फुफ्फुसीय उच्च रक्तचाप, हाइपरट्रिकोसिस, सिरदर्द, पेरिकार्डियल हैं बहाव आदि यह बालों के विकास का भी कारण बनता है इस प्रकार मिनोक्सिडिल सामयिक तैयारी (1-3%) खालित्य या गंजापन के इलाज के लिए उपयोग किया जाता है यह खालित्य के क्षेत्र में त्वचीय रक्त प्रवाह और रोम के बढ़ाव में वृद्धि का कारण बनता है
